摘要: Objective:To study the expression of aminopeptidase N (CD13) in renal carcinoma and the effect of CD13 expression vector transfection on biological behavior of cancer cells.Methods:Renal carcinoma tissue and normal kidney tissue were collected and APN (CD13) contents in tissue were detected; renal carcinoma cell lines kevt-3 were cultured, 0 μg/mL, 10 μg/mL, 20 μg/mL, 40 μg/mL and 80 μg/mL of CD13 expression vector were transfected, and then migration ability, ATP generation capacity, and mRNA contents of migration and angiogenesis genes in cells were detected.Results:mRNA contents of APN in renal carcinoma tissue were higher than those in normal kidney tissue; the higher the clinical stage and pathological grade were, the higher the mRNA contents of APN in renal carcinoma tissue were; mRNA levels of APN in renal carcinoma tissue with lymph node metastasis were higher than those without lymph node metastasis;CD13 expression vector transfection could dose-dependently increase kevt-3 cell migration rate, ATP generation amount as well as mRNA contents of VEGF, HIF-1α, MMP9 and MMP10.Conclusion: Expression of aminopeptidase N (CD13) in renal carcinoma tissue abnormally increases; overexpression of CD13 can promote renal carcinoma cell migration and increase ATP generation as well as VEGF, HIF-1, MMP9 and MMP10 expression.

  • Objective:To study the effect of preoperative compound rhizomacurcumaepowder treatment on serum markers and related signaling pathways function in endometrial tissue of patients with endometriosis.Methods:Endometriosis patients in our hospital were chosen for study and randomly divided into the treatment group (n=35) and the control group (n=35). The treatment group received preoperative compound rhizomacurcumaepowder treatment; the control group received preoperative conventional treatment. Then serum markers and function of related signaling pathways in endometrial tissues were compared.Results:After drug treatment, serum SCF, SDC-1, HLA-G, VEGF and Ang-2 contents of treatment group showed a significantly decreasing trend; mRNA contents and protein contents of Wnt, Frizzled, Dsh,β-catenin, cyclinD1, integrinα6β1, ERK1, ERK2, MMP9 and MMP27 in ectopic endometrial tissue of the treatment group all showed a significantly decreasing trend.Conclusion:Preoperative compound rhizomacurcumaepowder treatment is helpful to control the condition of endometriosis and inhibit the activation of cell cycle related signaling pathways and invasion related signaling pathways in ectopic endometrial tissue.

  • Objective:To study the miRNA-21 content in cervical cancer tissue and analyze its downstream target molecules.Methods:Patients with different FIGO stages of cervical cancer and healthy subjects were selected, cervical cancer tissue and normal cervical tissue were collected, and contents of miRNA-21 and apoptotic genes were detected; cervical cancer SiHa cells were cultured, miRNA-21 mimics and inhibitors were transfected, and then apoptotic gene contents were detected.Results:miRNA-21 contents in different stages of cervical cancer tissue were all higher than those in normal cervical tissue, mRNA contents of p16ink4a, ASPP1, Fas and GRIM-19 were lower than those in normal tissue, and mRNA contents of p16ink4a, ASPP1, Fas and GRIM-19 were negatively correlated with miRNA-21 contents; after miRNA-21 mimics were transfected, mRNA contents of p16ink4a, ASPP1, Fas and GRIM-19 significantly decreased, and after miRNA-21 inhibitors were transfected, mRNA contents of p16ink4a, ASPP1, Fas and GRIM-19 significantly increased.Conclusion:miRNA-21 contents in cervical cancer tissue significantly increase; downstream target genes of this miRNA may be apoptotic genes p16ink4a, ASPP1, Fas and GRIM-19.

  • Objective:To explore the impact of serum free amino acid and cytokines in patients with tuberculosis treated by Moxifloxacin.Methods:Chose 130 cases pulmonary tuberculosis patients,they were divided into observation group and control group according to random number table method, 65 cases in each group, all patients were given tuberculosis standard treatment, and on this basis, patients in control group were given levofloxacin tablets, patients in observation group were given moxifloxacin hydrochloride oral, they were treated for 6 months, compared the serum free amino acid and cytokine interleukin-18 (IL-18), tumor necrosis factor alpha-gamma (TNF-α), interferon (IFN-γ), interleukin-10 (IL-10) between two groups before and after treatment. The serum isoleucine, phenylalanine, threonine, leucine, valine, aspartic acid, glutamic acid and arginine in two groups after were increased significantly than before treatment, the difference was statistically significant, acquired aspartic acid, glutamic acid in observation group after treatment were significantly higher than the control group after treatment, the difference was statistically significant; The serum IL-18, IFN-γ, TNF-α and IL-10 in two groups after treatment were significantly reduced, the difference was statistically significant (P<0.05); the serum IL-18, IFN-γ, TNF-α and IL-10 in observation group after treatment were significantly lower than the control group after treatment, the difference was statistically significant.Conclusion:Compared with levofloxacin, moxifloxacin treatment tuberculosis can improve the patients' serum free amino acid levels, adjust the Th1/Th2 balance.

  • Objective:To study the modulation effect of Blu-ray irradiation combined with comprehensive treatment on serum indicators of neonates with hyperbilirubinemia caused by hemolytic disease of newborn. Methods:Eighty cases of neonates with hyperbilirubinemia caused by hemolytic disease of newborn were randomly divided into two groups, intervention group received Blu-ray irradiation combined with comprehensive medication and control group received comprehensive medication. Before and after treatment, contents of serum TBIL, MDA, nerve function related molecules as well as liver and kidney function related molecules were detected.Results:After treatment, serum TBIL, MDA, MBP, S-100β, NSE, Cr, BUN, CysC,β2-MG, αα-GST and GLDH levels of both groups were lower than those before treatment; serum TBIL, MDA, MBP, S-100β, NSE, Cr, BUN, CysC,β2-MG,α-GST and GLDH levels of intervention group were lower than those of control group.Conclusion:Blu-ray irradiation combined with comprehensive treatment can more effectively reduce bilirubin level, protect nerve function and reduce oxidative stress damage to liver and kidney function; it has positive value in treatment of neonates with hyperbilirubinemia caused by hemolytic disease of newborn.

  • Objective:To explore the expression change of serum and colon lavage fluid related indicators after probiotics combined with zinc preparations treatment of persistent diarrhea in children.Methods:80 cases of children with diarrhea treated in our hospital were selected. According to clinical symptoms, routine blood and stool test, they were all diagnosed with persistent diarrhea and randomly divided into control group and experimental group, 40 cases in each group. Control group received basic treatment and zinc preparations (zinc gluconate tablets) therapy; experimental group, based on treatment of control group, received probiotics (Siliankang) combined with zinc preparations treatment. Fasting serum and colon lavage fluid of both groups were collected before and after treatment to detect IL-6, IL-8, NO, MDA and SOD expression levels, and differences between two groups were compared.Results:After treatment, IL-6 and IL-8 expression levels in serum and colon lavage fluid of both groups decreased significantly than those before treatment, and those of experimental group decreased more significantly; after treatment, NO, MDA and SOD expression levels in serum and colon lavage fluid of both groups were significantly improved than those before treatment, and those of experimental group were closer to the normal levels than those of control group.Conclusions:Probiotics combined with zinc preparations treatment of persistent diarrhea in children has obvious curative effect; it effectively reduces expression levels of inflammatory cytokines and oxidative stress factors in serum and colon lavage fluid, and is worth popularization in future clinical treatment.

  • Objective:To study the vitamin D3 levels in primary Sjogren's syndrome patients and its correlation with severity and immune function.Methods:Active pSS patients, stable pSS patients and healthy volunteers were chosen for study. Peripheral blood was collected, mononuclear cells were isolated and percentages of CD27high plasma cells and CD27+ memory B cells were detected; serum was collected and contents of 1,25(OH)2D3, TNF-α, IL-6, IL-10 and IL-17 were detected.Results:(1) 1,25(OH)2D3 contents in serum as well as CD27high plasma cell and CD27+ memory B cell contents in peripheral blood of pSS group were lower than those of control group; serum TNF-α, IL-6 and IL-17 contents were higher than those of control group and IL-10 contents were lower than those of control group; (2) 1,25(OH)2D3 contents in serum as well as CD27high plasma cell and CD27+ memory B cell contents in peripheral blood of active pSS patients were lower than those of stable pSS patients; serum TNF-α, IL-6 and IL-17 contents were higher than those of stable pSS patients and IL-10 contents were lower than those of stable pSS patients; (3) 1,25(OH)2D3 level was positively correlated with contents of CD27highplasma cells, CD27+ memory B cells and IL-10, and negatively correlated with TNF-α, IL-6 and IL-17 contents.Conclusion:Abnormal reduction of Vitamin D3 levels is involved in the pathogenesis of primary Sjogren's syndrome and closely related to the severity and immune function of the disease.

  • Objective:To explore the effect of percutaneous vertebroplasty (PVP) in the treatment of osteoporotic vertebral compression fracture (OVCF).Methods: OVCF patients who were admitted in our hospital from April, 2014 to April, 2015 were included in the study and randomized into the observation group (PVP treatment) and the control group (conventional treatment). The patients in the two groups were given freeze-dried powder of Xuesaitong, Xianling Gubao Capsules, and Caltrate. On this basis, the patients in the observation group were given PVP. After anesthesia, in the guiding of X-ray normal perspective, puncturation to the anterior 1/3 place of the affected vertebra through the vertebral pedicle was performed, and bone cement was infused. The patients in the control group were given hard bed and drugs for analgesia and sedation. A restoration cushion was placed in the affected vertebra and the height was adjusted according to the tolerance degree. Those who were relieved after 3 d were given functional exercise by the method of 5 point support hance.Results:VAS score after 1-week treatment in the observation group was significantly lower than that in the control group. The comparison of VAS scores after 1-month and 3-month treatments between the two groups was not statistically significant. The mobility score after 1-week treatment in the observation group was significantly lower than that in the control group. The comparison of mobility score after 1-month and 3-month treatments between the two groups was not statistically significant. After 1-month treatment in the two groups, the Cobb’s angle was significantly reduced and the vertebral height was significantly elevated when compared with before treatment. The comparison of Cobb’s angle and vertebral height after 1-month treatment between the two groups was not statistically significant.Conclusions: PVP has the advantages of short course, and rapid pain relief and function recovery, and its long-term effect is not significantly different from that by the conventional treatment. Meanwhile, PVP has a possibility to induce the leakage of bone cement; therefore, the indications should be rationally mastered in the clinic.

  • Objective:To explore the influence of inflammation factors and intestinal mucosal barrier function on severe acute pancreatitis patients treated with bifidobacterium triple viable capsules.Methods:88 cases severe acute pancreatitis patients were divided into observation group and control group according to admission order, 44 cases in each group, all patients were given conventional treatment, on this base, patients in observation group were treated by oral bifidobacterium triple viable capsules, they were treated for one week, detected the serum inflammatory factors: the tumor necrosis factor alpha (TNF-α), interleukin 6 (IL-6) and C-reactive protein (CRP) and intestinal mucosal barrier function index: endotoxin, diamine oxidase (DAO), D-lactic acid, urinary lactulose/mannitol (L/M) in the two groups before and after treatment.Results:The levels of TNF-α, IL-6 and CRP in two groups after treatment were significantly reduced than before treatment, and after treatment the levels of TNF-α, IL-6 and CRP in observation group was significantly lower than that of the control group, all the difference was statistically significant; The levels of endotoxin, DA, D-lactic acid, L/M in two groups after treatment were significantly reduced than before treatment, and the levels of endotoxin, DA, D-lactic acid,L/M after treatment in observation group was significantly lower than that of the control group, the difference was statistically significant.Conclusion: Based on conventional treatment combined bifidobacterium triple viable capsules can significantly reduce inflammation in patients with severe acute pancreatitis, it can protect the intestinal mucosal barrier function, and has important clinical significance.

  • Objective:To explore the effects of early enteral nutrition combined with triple viable capsule of bacillus on inflammatory factors and intestinal mucosal barrier function in treatment of severe acute pancreatitis.Methods:118 cases of severe acute pancreatitis were randomly divided into two groups. The control group (59 cases) patients were treated with early enteral nutrition therapy, while the observation group (59 cases) was treated with early enteral nutrition combined with triple viable capsule of bacillus. The clinical efficacy of the two groups and the changes of the levels of inflammatory factors and the function of intestinal mucosal barrier were compared.Results:The time of abdominal pain disappearance, the abdominal distension disappeared, and the recovery time of the exhaust function of the observation group were (2.30±0.30) d, (3.39±0.51) d and (2.68±0.78) d, were significantly decreased compared with the control group (P<0.05). IL-6, IL-8, TNF-α and CRP of observation group were (33.19±355) g/L, (30.28±3.71) g/L, (36.33±3.53) g/L and (12.19±1.18) mg/L, were significantly lower compared with those before treatment (P<0.05) and was significantly lower than that of the control group (P<0.05). After treatment, D-lactate, DAO, endotoxin and exami of observation group were (544±0.54) mg/L, Dao (4.11±0.34) U/L, (0.65±0.18) EU/L, (0.15±0.07), were decreased significantly compared with those before treatment (P<0.05) and was significantly lower than that of the control group (P<0.05).Conclusion:Eearly enteral nutrition combined with triple viable capsule of bacillus on inflammatory factors and intestinal mucosal barrier function in treatment of SAP has significant effect, can rapidly improve the symptoms of patients, reduce inflammation, promote the recovery of intestinal mucosal barrier function, it is worth clinical reference and popularization and application.

  • Objective:To explore the effect of caffeine citrate on the blood gas indexes in newborns with apnea.Methods: A total of 108 primary premature infants diagnosed with apnea were included in the study and randomized into the caffeine citrate treatment group and the aminophylline control group with 54 cases in each group. The apnea episode duration 72 h after medication, and the blood gas before medication and 24, 48, 72 h after medication between the two groups were compared.Results: The apnea episode duration 24, 48, 72 h after medication in the treatment group was significantly shorter than that in the control group. PaO2 24, 48, 72 h after medication in the treatment group was significantly higher than that in the control group, while PaCO2 was significantly reduced when compared with the control group.Conclusions:Caffeine citrate has achieved a significantly clinical effect in the treatment of newborn apnea, as a new therapeutic drug, is worthy of being widely recommended in the clinic.
