    Objective:To study effects of the exogenous bone mesenchymalstem cells ( BMSCs) transplanta-tion on silicosis fibrosisin rats, and to explore dose-effect relationship. Methods:BMSCs were isolated and cul-tured from male 5-week-old SD rats in vitro. Fifty healthy female SD rats were randomly divided into five groups:model control group, silicosis model group, BMSCs treatment A group (1x106/ ml), BMSCs treatment B group (3 x106/ ml) , BMSCs treatment C group (5x106/ ml)(n=10). The silicosis model were made by One-time infu-sion of silica dust suspension, using the non-exposed tracheal intubation, and given different doses of BMSCs as intervention therapy. All the rats were sacrificed on the 21st day after model estalishment. The morphological chan-ges of lung tissue were observed by HE staining and Masson staining; the localization and distribution of TNF-αand TGF-β were detected by immunohistochemistry; the expression quantity of TNF-α, TGF-β, collagen I and collagen III protein were detected by Western blotting;the sex-determining gene SRY was searched by immunoflu-orescence to confirme BMSCs home to damage lung tissue. Results: Compared with the model control group, the silicosis model group had significantly alveolitis changes, silicon nodule formation, collagen deposition and other pathological changes. Compared with silicosis model group, the pathological changes above in BMSCs treatment A group were improved;the condition of BMSCs treatment B group was improved significantly;the pathological chan-ges of BMSCs treatment C group were increased obviously. the expression of TNF-α, TGF-β, collagen I and col-lagen III protein in lung tissue:BMSCs treatment C group>silicosis model group>BMSCs treatment A group>BM-SCs treatment B group>model control group. The difference of BMSCs treatment C group and silicosis model group had no statistically significant ( P>0 . 05 ) , and the difference between the other groups had statistically significant 158 (P<0. 05). The SRY-positive cells were observed in BMSCs treatment B group by laser scanning confocal microscope, but no significant expression in the heart, liver, spleen and kidney tissues. Conclusions: The Exogenous BMSCs transplantation antagonize rat silicosis fibrosis, which has dose-effect relationship. Our data suggested that the best transplantation dose for silicosis fibrosis is 3x106 / ml.

  • 硫化氢对糖尿病大鼠心肌纤维化及MAPK1/3和MMP-8表达的影响

    目的:探讨气体信号分子硫化氢(H2S)对糖尿病大鼠心肌纤维化及MAPK1/3和MMP-8蛋白表达的影响。方法40只SD大鼠随机分为4组:正常对照组、糖尿病大鼠组(STZ组)、硫化氢干预糖尿病大鼠组(STZ+H2S组)和硫化氢干预正常大鼠组(H2S组)。用腹腔注射链脲佐菌素(STZ)(40 mg/kg)建立糖尿病大鼠模型;对照组每天腹腔注射生理盐水;STZ+H2S组与H2S组每天腹腔注射硫氢化钠(H2S供体)溶液(100μmol/kg),8周后处死大鼠,HE染色观察心肌纤维病理形态学变化,Western blot法检测Collagen I、MAPK1/3和MMP-8蛋白的表达。结果与对照组相比,糖尿病组大鼠心肌组织胶原含量明显升高,心肌间质纤维化,Collagen I、MAPK1/3和MMP-8蛋白表达水平显著升高(P<0.05);糖尿病大鼠经硫化氢干预后大部分心肌纤维呈平行排列,结构清晰,间质有少量疏松结缔组织,Collagen I、MAPK1/3和MMP-8蛋白表达水平明显下调(P<0.05),而非糖尿病硫化氢干预组心肌纤维化程度及心肌组织Collagen I、MAPK1/3和MMP-8蛋白表达水平较对照组均无明显差异(P>0.05)。结论 H2S可改善糖尿病大鼠心肌纤维化,其机制可能与抑制MAPK1/3/MMP-8信号通路有关。

  • CUEDC2对梗阻性肾病小鼠肾组织炎症反应及间质纤维化的作用

    目的:研究CUEDC2对单侧输尿管梗阻(UUO)小鼠肾组织炎症反应及间质纤维化的作用。方法:Balb/c 小鼠30只,随机分为假手术组、UUO组和CUEDC2治疗组。 HE 及Masson 染色观察肾脏病理改变;ELISA法检测炎症因子表达;免疫组化法检测肾组织CUEDC2表达;Western Blot检测CUEDC2、纤连蛋白(Fibronectin)、钙黏蛋白(E-cadherin)、胶原蛋白I型(Collagen I)表达。结果:术后7、14 d,CUEDC2治疗组的肾间质纤维化相对面积、炎症因子 ICAM1、MCP1、IL-1、IL-8表达及 Fibronectin、Collagen I 表达均较 UUO 组明显减少(P <0.05),E-cadherin 表达明显增加(P <0.05)。结论: CUEDC2可以抑制多种炎症因子表达,减少间质胶原沉积,减轻肾脏纤维化。
