ELISA双抗体夹心法检测急性肝炎患者抗-HAV-IgM及其动态变化的研究
用人胚肺二倍体细胞培养的甲型肝炎病毒作抗原,以市售马抗人IgM经纯化后的抗体为捕获抗体,以纯化甲型肝炎恢复期血清为特异性抗体,过碘酸钠法标记酶,以PPD为底物制成抗一HAV-IgM诊断试剂盒,并建立ELISA双抗体夹心法检测抗-HAV-IgM,其结果:甲型肝炎暴发点44例患者均为阳性,流行性出血热30例均为阴性,100例急性病毒性肝炎住院患者中30例阳性.对一例甲型肝炎患者作了抗-HAV—IgM动态观察,发病前40天为阴性,而病后一周及一个月其滴度达10-7以上,2个月后滴度明显下降,病后4个月为阴性.

乙型肝炎患者HBsAg携带者血清中多肽图谱的研究
本文建立了自少量血清中研究I-IBsAg多肽图谱的方法,患者血清经纯化、SD.S聚丙烯酰胺凝胶电泳及Western吸印后,HBgAg多肽用酶免疫染色或放射自显影进行分析,发现10例慢性乙肝患者血清HBsAg多肽组成有明显差别.同一来源的HBV毒株在不同个体或同一个体随病程不同多肽图谱亦不同.血清HBV DNA阳性与多肽P39/P42的关系较P31/P34为密切,提示Pre-S1抗原不仅在急性感染中与病毒复制有关,在慢性感染中亦有关,PHSA'受体与P31/P42的符合率为94.1%,反映这一多肽确带有PHSA受体.

Percutaneous ethanol injection therapy(PElT) has been established as an interventional method of the ablation of hepatic malignancies, especially hepatocellular carcinoma(HCC), since long. Its effects are based on cellular dehydration,toxic and coagulation necrosis and thrombosis of small venules within the tumour. The result is fibrosis of the damaged tissue, loss of function and shrinkage. Hence the "ablative" effect of absolute(95%) ethanol. The more confined ally lesion the better the ablative effect due to the high concentration of the toxic liquid. Highest effects will be achieved in encapsulated nodules which prevent diffusion of the toxic agent into surrounding normal tissue which is the case in HCC.

为推广和提高Ilizarov技术在骨科的应用而努力
1981年,Ilizarov的理论与技术传入到意大利,1982年在意大利成立第一个Ilizarov方法研究与应用学会 (Association for the Study and Application of the Method of Ilizarov,ASAMI ),对这项技术进行了全面的引进、学习、研究与临床应用.1986年Ilizarov的理论与技术传入到北美,由美国马里兰大学骨科系Dror Paley教授于1988年发起并成立ASAMI学会,并召开了多次世界性的学术会议,仅在美国Baltimore市就连续举办了16届肢体畸形国际培训班.1989年美国临床骨科和相关研究杂志(J Clin Orthop)连续2期刊载Ilizarov于80年代前即完成的2篇动物实验研究论文,牵张-应力效应对组织生长及起源作用的实验研究:(1)软组织保留和固定的稳定性对骨折愈合的影响;(2)牵引频率和速度对骨愈合的影响.

儿童尿石透射电镜的初步观察
本文将11例儿童术后的尿石在2700~6000倍透射电镜下观察,根据摄象影象将尿石分为致密型板状结构、致密型粒状结构、疏松型混合结构、杂乱型晶状结构.提出了除原始核心以外还存在有多个卫星型核心的观点,指出尿石的疏密程度除与尿液的化学成分有关外,还与其吸附的能量有关.

“DC细胞工厂”--脐带血CD34+造血干细胞来源的DC的大量制备
背景:树突状细胞(Dendritic Cell,DC)充当着免疫系统中的“警察”角色,它能够识别体内异常抗原信息并通过激活初始T细胞而诱导特异性的免疫应答。以DC为基础制备的肿瘤疫苗能有效的构筑抗肿瘤免疫屏障,因此DC被认为是充满前景的免疫治疗工具。作为原料细胞,大量成熟,功能均一的DC 细胞是制备DC疫苗的必须条件,而临床上广泛应用的DC细胞来自于自体外周血,数量在106~108之间,无法充分满足临床应用。因此,探索新的大量制备DC的方法成为临床上迫切的需求。目的:本实验旨在探索新的由CD34+造血干细胞大量诱导DC细胞生成的方法,并通过体外实验证实这种CD34-DC肿瘤疫苗相比外周血单核细胞来源的DC疫苗(PBMC-DC)具有类似的或者更好的诱导抗肿瘤免疫应答的能力。方法:分离脐带血CD34+造血干细胞并利用GM-CSF/SCF进行连续30 d的扩增培养,期间通过贴壁持续收获DC。外周血单核细胞诱导的DC作为对照。对比两种来源的DC形态,表型和包括抗原吞噬,混合淋巴增殖,细胞因子分泌,趋化能力的功能特征。分别将其制备成肿瘤DC疫苗,并对比体外诱导的CTL IFN-γ分泌能力和针对肿瘤靶细胞的细胞毒性。结果:1) GM-CSF/SCF细胞因子组合连续培养CD34+细胞30 d后,共收获~1010总细胞量和~109的DC。与PBMC-DC相比,具有类似的形态特征;2)成熟状态时, CD34-DC与PBMC-DC具有类似的细胞表型(CD80, CD83,CD86,HLA-DR),而未成熟时CD34-DC表型好于PBMC-DC,尤其是CD80表达较早上调;3)无论是新鲜的还是冻存后复苏的,CD34-DC和PMBC-DC在GM-CSF/IL-4培养的第5天吞噬功能强,是抗原负载的佳时间窗;4)鸡尾酒式细胞因子作用下, CD34-DC较PBMC-DC具有更强的IL-12p70和IL-10的分泌能力,且前者诱导的CTL IFN-γ分泌水平更高,同时5)其表现出了更强的细胞毒作用;6)正常健康孕妇脐带血中CD34+造血干细胞具有稳定的增殖和向DC分化的潜能。结论:我们建立了一套由脐带血造血干细胞诱导大量DC生成的方法并称之为“DC工厂”。由其制备的DC抗肿瘤疫苗是将来能够用于临床的充满前景的免疫刺激剂。

Elemene is a new anticancer drug isolated from a Chinese traditional medicine Curcuma aromatica. In previous work, we discovered that tumor cell vaccine (TCV) treated with oleum Curcuma aromatica or elemene could induce significant immunoprophylactic effect against a variety of aminal tumor strains and the method of preparation of elemene combo-TCV(EC-TCV) already got China's inventive patent. In this paper we further studied the active immunotherapeutic effect and the possible cellular/molecular mechanisms of EC-TCV immunization. The results were as follows:(1) EC-TCV immunization showed significant therapeutic effects (P<0.05) against murine Ca761 syngeneic mammary carcinoma (H-2k) and HCa-F allogeneic hepatic carcinoma (H-2-) models; (2) The spleen cells of Hca-F EC-TCV immunized mice displayed higher cytotoxicity and IL-12 level while the secretion of IL-10 was decreased (P<0.05); (3) Similar to heat shock, elemene(E), mitomycin C(MMC) and glutaraldehyde (G) could act alone as stressor, and induce significant changes of the expression of membrane heat shock proteins(HSP70 or/and HSP90) on L615 leukemia and HCa-F hepatoma cells and the EC-TCVs (E+MMC+G treated in combination) showed the highest level of membrane HSPs expression (P<0.05 or P<0.01 );(4) The HSP70-peptide complex isolated from HCa-F EC-TCV through ADP-agarose affinity chromatographic system could induce active immunoprotection against lethal dose challenge of HCa-F hepatic cancer cell but could not protect against the cross challenge of lethal dose of L615 leukemia. The results indicated that the immunoprotective effect of EC-TCV was in some extent tumor-specific, MHC-nonrestricted, and HSPs might play an important role in its molecular mechanisms.

AIM: The aim of this study was to explore the expression of nerve growth factor(NGF) in spinal dorsal horn following crushed spinal cord injury. METHODS: The adult Srague-Dawley rat model of crushed spinal cord injury was established by the method in our laboratory, and intact spinal cord was used as control. The rats were sacrificed respectively after 24 hours, 7 days, and 21 days of operation, and the L3 spinal segments were removed out and fixed in 4% polyformaldehyde. The segments were sectioned into sections of 20 μm in thickness. The sections were stained with anti-NGF antibody by ABC method of immunohistochemistry technique. The immunoreactive intensity of NGF and the number of positive neurons as well as glial cells in dorsal horn were observed and counted under light microscope. RESULTS: The number of positive cells and immunoreactive intensity of NGF increased gradually in the dorsal horn at 24 hours, 7 days and 21 days following crushed spinal cord injury compared with control group (P<0.01). CONCLUSION: These results indicated that NGF plays an important role in the postoperative reaction during the early period of the crushed spinal cord injury.

毛囊干细胞及其分离与识别方法的研究进展
干细胞是指未成熟细胞,具有自我更新和分化能力,可分化为胚胎干细胞和成体干细胞、其他细胞、组织及器官[1],胚胎十细胞是有应用价值的干细胞,但由于来源于胚胎,进行相关研究和治疗必然涉及伦理和道德问题,所以胚胎干细胞的研究受到限制,而来源于同体的成体干细胞就不存在这种限制.皮肤组织更新速度快、再生能力强,因此,来自皮肤局部的干细胞受到了重视.
